Am I right to say that the safest way to do it is with a shuttle like a Leopard, no autopilot? And I’m guaranteed to reach my destination in one piece? Perhaps even use my capsule to make a test run first to check for gate camp? Any other precaution?

sunesis can reach 30k EHP with <2s align.

Leopards may be fast, but they aren’t safe or immune to getting killed. Not even in high sec.
For example, this guy kills them often:
You’ll never bee ‘guaranteed to reach your destination in one piece’ in EVE, but you can reduce the risk a lot.
The best way to reduce the risk of transportation is to pay someone else to take that risk, with a courier contract with the right amount of collateral.
If you wish to move the blueprint yourself, look into a fast aligning ship (below 2 or even 1 second align time) with high amount of hitpoints (not a shuttle) and avoid obvious routes such as the direct warp from Jita station to the Perimeter gate to avoid getting smartbombed.

Gate campers often use these to can lock you faster, fast enough so you can’t escape them.
One option for a sub 2 sec align but well tanked ship is the Sunesis:
Though for this task you should active tank it in case you expect to run into a smartbomb gatecamp (low and nullsec) and best to have a scout check right before you would jump to see what’s on the other side and at the exit gate as well.
And returning to the subject of sensor boosters also keep in mind anything (like shield extenders for example) that increase your signature radius will make you easier (and quicker) to lock and with enough sensor boosters applied they might even lock you if you fit for sub 2 sec align if I understand the mechanics correctly. So active tanking or adding more shield strength will help you against smartbombs but will make you more vulnerable against sensor boosted target lockers.
If you attempt this on your own but even if you contract someone to do it for you be sure to link is the lossmail if that is how it ends, sounds like a juicy fun killboard material to witness.

The Slasher didn’t make it obviously but the Sunesis fit for sub 2 sec align plus kitted with shield extenders plus two multispectrum shield hardeners did the job flawlessly.
I think I was able to tank at least four of them and did the passage like six times back and forth. With the active tank and omniresists of the Sunesis they barely did damage.
However keep in mind the additional signature radius of the shield modules might make you much easier to catch with sensor boosted camps and also you need your shield hardeners active when you get hit by the smartbombs otherwise your resists will be much lower. Having lag spikes and mishaps can prove lethal pretty quick.

So if I count correctly it was five times and three of them though if I recall correctly there was plenty of buffer left to withstand more.
Also note that this is a lowly trained Alpha character so with highly trained Omega skills it should perform even better.
I think the SB in this case referred to smartbombs, which are the biggest danger for shuttles as it only takes one cruiser with smartbombs at the wrong time and wrong place to get your shuttle killed.

Ah right. Well my posts help with and cover both subjects at the same time. 
Best way to move it is to not tell anyone you are moving it and from where to where.
But usually under 2 sec align time with an insta undock bookmark 2000km off station and a 0 book mark on the other station.
Also blockade runners are immune to cargo scanners.
If you try move it yourself now that you told everyone, someone will sit in a smart bomb bs with safety set to red on the gate and wait for you to fly passed and purposely concord themselves with smart bombs to kill any leopard you have.

Some people specialise in killing shuttles (and pods) using a smart bomb ship on gates. A well timed SB will hit you while you’re still in warp, just as you’re landing on gate, so your align speed doesn’t really matter.
Double shrink wrapped courier contract in max tank bowhead filled with as many T1 worthless ships as you can fit. Needle in a haystack.
Also directly in warp. There is a guy that uses cruiser with SB to kill shuttles, especially Leopards, mid warp to jita. Cloak also don’t help. So afik only way to avoid SB damage is to not be in range. Best way to achieve this is to travel using bookmarks at gates and stations. Which helps to warp in and land at different angels than normal trade route.
Fun fact: You cannot smartbomb directly at gates or wormholes. So reaching the gate by a different angle (“bouncing” from a celestial, station, or bookmark) can save your ship.
